Study of Electrophoresis of Isozymes and Soluble Proteins of Diploid Perennial T eosinte and Its Hybrid F1 with Maize

Abstract

Using polyacrylamide gel electrophoresis we analysed the zymogram patterns of pe roxidase isozymes for 16 hybridized combinations of diploid perennial teosinte × maize hybrid F1.The results showed that the enzyme bands of interspecific hyb rid F1 mainly displayed complementary pattern of two parents.According to our da ta,they could be divided into three zymogram patterns:(1) Completely complementa ry band pattern;(2) Sectionally complementary bands pattern;(3) Hybrid band patt ern.We also found that the hybrid enzyme bands often appeared in the region vary ing from Rf 0.21 to 0.29,but the enzyme bands disappeared in the region from Rf 0.21 to 0.40.During the experiments,hybrid enzyme bands of alcohol dehydrogenase had never been observed on all combinations examined. The results obtained from soluble protein electrophoretogram scanning sugges ted that the protein molecules of diploid perennial teosinte possessed a special marked protein peak (Rf 0.28) which was different from that in maize.
